The isolation separates PBMCs from other components in the blood, enabling their use in research. PBMCs are part of a network of cells responsible for immune response against pathogens and cancerous cells.

ELISpot, FluoroSpot and Flow cytometry assays are commonly used to assess cellular immunogenicity at a single-cell level by measuring a PBMC sample’s ability to produce and secrete cytokines in response to antigen stimulation.

PBMCs are isolated from freshly collected blood. Depending on the quality (viability, recovery) of PBMC required for downstream analysis, the TAT between fresh blood sample collection and PBMC isolation may vary greatly. In recent years downstream analysis often requires rapid PBMC isolation, such as within 8-10 hours.

To perform PBMC isolation within a short timeframe (e.g., <8 hours), the PBMC processing laboratory must be closely located (in a driving distance) to the Clinical site. This can be achieved by two ways:

Local PBMC processing labs are selected, trained, and qualified for the task

As an exception: Clinical sites are trained to perform the PBMC isolation

Cerba Research will require anywhere between 2-6 months to qualify a new PBMC processing laboratory. Depending on the PBMC isolation protocol, location, specific requirements. The lead time to qualify a lab that is already identified and operational in our network can be significantly less.
