This study presents an optimized and scalable workflow for recovering high-quality, intact genomic DNA from lymphodepleted clinical samples, a critical step for reliable CAR-T vector analysis. By improving DNA extraction from low-cellularity blood and bone marrow samples collected after lymphodepletion, the method ensures sufficient yield and integrity for downstream assays such as vector copy number (VCN) and replication-competent virus monitoring. The approach enhances the robustness and consistency of CAR-T therapy monitoring during the early post-infusion period, when sample material is limited.
